THE ALKALOIDS OF RAUWOLFIA YUNNANENSIS TSIANG

Abstract: Four alkaloids had been isolated from the roots of Rauwolfia yunnanensis Tsiang. Three of them are identified as reserpine, ajmaline and ajmalicine res-pectively by comparing their physical and chemical properties with those of the authentic samples. It seems that the other remained alkaloid is a new compound which has the formula C22H26O4N2 on the elementary analysis of base and it's hydrochloride salf, [α]D+l06' (CHCl3). It should be decomposed by heating without characteristic meltinq point. According to it's IR spectrum it has indole group, hydroxyI group and αβunsaturafed carboxylic metbyl ester and seems to be a serpentine derivative.

摘要: 云南萝芙木(Rauwolfia gunnanensis Tsiang)为国产成药“降压灵”的原料,魏均娴曾对其化学成份进行过研究,用氧化铝层析法分离得利血平和Ajmalicine, 后未见其他报道。我们在研究利血平的植物资源和利血平的生产工艺时曾从云南萝芙木分离出四种生物硷,一为微黄色针晶,熔点264—5度 ,[a]D-119度(CHCl3),Frobde反应:(黄)→绿→兰,香草醛反应:玫瑰红色;对二甲氨基苯甲醛反应:兰绿色,加冰醋酸后转为血红色;K—K反应:界面呈兰色,浓硝酸反应:(兰绿)→红棕。其盐酸盐熔点222—4℃(分解),其硫酸盐熔点261—2℃(分解),其硫酸盐熔点245℃ (分解),UVEtoHmax 218nm(logε4.85),267nm(logε4.39),297nm(logε4.20),UVmin246nm(logε 4.12),287nm(log ε4.21),以上数据与利血平吻合,在氧化铝上作薄层层析,氯仿亿醚(7:3)展开,与已知利血平对照出值相同,其IR与Sadler标准图谱基本一致,因此鉴定为利血平。二为无色针晶,熔点259℃(分解),[a]D-39.6℃(吡啶),其盐酸盐熔点268℃(分解),其硝酸盐熔点258℃(分解),在上法薄层层析上与已知Amalicine的Rf值一致,根据红外光谱,鉴定为Ajmalicine。
